If you let it sit, overnight, all you will lose is the oil that is in the filter, and what's still in the engine, if your filter has a backflow valve. You should never pour fluid from a filter back into the system it came from.

You should change the filter at the regular maintenance interval. Synth is highly resistant to thermal breakdown, which is why it lasts so long, but it doesn't do anything to prevent contamination. It's also not a bad idea, if you are running synth for extended intervals, to get the oil checked periodically.

Personally, I just run synth and change fluid/filter at 7500mile intervals.
