The fuel vapors from the fuel tank collect in the charcoal canister. At cruising speed, with a low load on the motor, the purge valve will open and the motor will suck the vapors from the canister. If you leave the line off long enough the ECU might see a leak in the purge system, and you might get a check engine light on. Make sure you plug the end going to the intake manifold or you'll have a pretty good vaccum leak.
